A mechanic advised me that coil #3 was not firing in my car, can this cause it to burn more gas than it normally does?

You mean one of the six cylinders is not producing power so the other five have to work harder? And cylinder number 3 is still pumping gas but not burning it so it is just wasted and going out the tail pipe? You bet that will affect fuel mileage.
