Hi,
No need to check for spark. If it started with starting fluid, spark is good. Fuel is the issue.
Do me a favor, if you haven't already, make sure fuel pressure is within the manufacturer's specs at the fuel rail. Here is a link that shows how it's done:

Here is the specification information:
2000 GMC Truck K 1500 Truck 4WD V8-5.3L VIN T
Fuel Pump Pressure
Vehicle Powertrain Management Fuel Delivery and Air Induction Fuel Pump Fuel Pressure Specifications Pressure, Vacuum and Temperature Fuel Pump Pressure
FUEL PUMP PRESSURE
Fuel Pressure
Key On Engine Off 55 - 62 psi
I realize you replaced the pump and other components, but if the pressure is too low, it won't run.
If you find the pressure is good, then we need to check for an injector pulse at the fuel injectors. Something is preventing the fuel from getting into the engine. Here is a link that explains how to check the injectors:

Don't worry about the ignition spark at this point. I know it's working based on your test.
Let me know what you find or if you have other questions.
Take care,
Joe
