Hello again,
No, one spark plug keeping a cylinder from firing shouldn't cause the engine to not start. Is your Malfunction Indicator Lamp (MIL) or "check engine" lamp illuminated? Also you have fuel pressure as you can here the fuel pump turn on and prime the fuel system, but are you getting fuel injector pulse to turn the fuel injectors on and off? In the link below we have included a guide describing how to easily test your fuel injectors to make sure the Power-train Control Module is providing the proper duty cycle for each fuel injector.

Another way to test individual fuel injectors that are out of the vehicles on the is to securely fix a hose on the inlet of injector with a spray can of carburetor or brake cleaner spraying through the straw, which in turn, is attached securely to the hose. Get two electrical connector pins that fit on the fuel injector pins and hook up to a 9 volt battery with two wires. The 9 volt battery will provide enough current to open the fuel injector and ensure that the internal solenoid is working as it is supposed to. Spray the carburetor/brake cleaner and as the pressure builds it will get sprayed by the energized fuel injector. Please get back to us with what you are able to find out.
Thanks,
Alex
