When I start my truck in the morning and go to the school, when I pull out I accelerate and I heard a low pop in the front and it start to shake when I press the gas. And then I went to a dealer to do a scan and he said cylinder 5 is "missing" accordingly to the scanner. So I tell him all the stuff I did and he said its backfire, it usually in the back. Then the next day we change all the spark plugs, switch around the ignition coil, we crank it up, drove it down the block. And its better but it still shake and the engine light start to blink.

The shaking you described is indeed due to a misfire, and the diagnostic fault code says it's cylinder number five, but when you say,

"So I tell him all the stuff I did",

you have to explain that if it's relevant. There's way too many possibilities for me to guess what you did. The next step after new spark plugs is to swap ignition coils, but you also have to erase the fault code, then see if it sets again for the same cylinder or the one you switched that ignition coil to. If the new code is still for cylinder number five, swap two injectors and try again. Also consider performing a compression test.

When the Check Engine light is flashing, you're supposed to stop the engine right away. It's telling you too much unburned fuel is going into the exhaust system and it's going to overheat and damage the expensive catalytic converter.
