Hi and thanks for using 2CarPros. Com.
To properly check the transmission fluid on your vehicle, the engine should be at operating temperature and running. Open the hood and locate the transmission dipstick. It is bright yellow of orange in color. It will be located on the transmission front slightly back from the transmission bell housing. I have attached a picture of a fill tube for you to see. That is where the dipstick should be.

If you find fluid needs to be added, make sure to get fluid which meets the manufacturers recommendations.

FLUID TYPE SPECIFICATIONS
Fluid type: Toyota Genuine ATF Type T-IV
Change Automatic Transmission Fluid Only As Necessary.

Use only "Toyota Genuine ATF Type T-IV" (ATF JWS3309 or NWS6500) to aid in assuring optimum transmission performance.

I hope this helps. Let me know if you have other questions.

Take care,
Joe

Is it important to let car idle for a minute or so and put "gear selector" in each selection ending in "park" before checking fluid level?

Hi. No just make sure it is in park. When you service a transmission, it is a good idea to run it through the gears, but just to check level, it is not necessary.

Let me know if you have other questions.
