Transmission fluid check, do I check it when the vehicle is running?

My understanding is that you keep the car running until it's optimum temperature then check the dipstick while car is running, correct?

You are correct, you check the transmission fluid with the engine/trans fully warmed up. Attached is an image of the written service procedure for your reference.
