I have to replace the whole coolant line, and cannot get the end that is attached to the transmission off.

Those are push connect fittings, you slip the tool down over the line and they should pull free. Then you sort of pull the line down and out. Make sure you have the front out first as the line needs to twist a bit for it to come out.
