I had no heat because the heater core was plugged up so I unplugged the PCM so I would have room to flush the heater core. After I flushed the heater core and plugged in the PCM I started the car the transmission caution light is flashing and it will not shift to overdrive. What have I done?

it could have bent a pin in PCM or something like that. have It scanned for codes to see if any are on and get back to us with the code. if you unplug it again look for a bent pin then straighten it and carefully plug the PCM back in and use dielectric grease for better contact.
