1995 Pontiac Transport intake manifold

I am going to replace the upper intake manifold gasget on my transport (3.8).I noticed a coupling on the fuel line configuration that appears to be for pressurizing.Will the lines have to be pressurized some way after repair?

No, the fuel pump will pressurize it as soon as the key is turned on.
