If you have a miss on only one cylinder you start with testing the ignition first because it is generally easier. However other than testing for power to the coil and the ground a regular meter is useless. For primary and secondary voltage testing you need an amp clamp and a scope.
Testing of the injection side is about the same, but a noid light on the injector control wire would tell you if it was being fired, it will not show you if the pattern is any good but at least you can see if the controls are working.
