Random/multiple cylinder misfire

Typically on misfires the you will get a trouble code.
Is your check engine light illuminated? If so what codes are you getting?

Have you got a chance to check the ignition control module?
The ICM controls the coil and plugs. A bad ICM can produce random misfires.
The ICM can be tested, usually you can take it off and take to your local auto parts store and they will test it for you. This way if it is not faulty you just don't want to replace it.

Does the EGR valve or throttle positioning sensor have anything to do with misfire also?

If you have a clogged EGR or a dirty TPS it can run rough, but should not trigger misfire.
Typically you will have idling issue, like rough idle or low power.

I also hear a knocking noise like it could be a lifter.

Lifter noise can be heard either for being low on oil or an issue with the lifter.
Sometimes engine misfire can create knocking noise. Did you get a chance to scan for codes?

Code says random/multiple cylinder misfire.

The random misfire can be triggered by ignition control module. If you have not already might want to get that checked. That module is right next to the coil.

I haven't checked it yet. but I would like to know if the spyder injectors might cause the misfire?

Well, the spyder injector can cause the misfire. Used to see those injectors getting changed all the time under warranty due to all sorts of issue.
But typically the replacement ones held up better.

Changed ICM still misfires and knocks not saying bad coil but coil is new.

You know the other item could be the distributor itself. Do a thorough inspection of the distributor. See the guide below, it does over the inspection process.

If it's not the distributor, do you suggest changing the spyder?

Well,
if you have checked all ignition components like plugs, wires, cap/rot, distributor etc.
Then the spider injector really is the next item. But keep in mind many times those random misfires are due to an issue with the ECM.

I've changed wires,plugs,cap,rotor bitton,coil,icm,mass airflow The only thing left is the distributor and spyder.

I have seen many of these spyder injectors cause misfire and many other drivability issue back when they first came out. The replacement ones typically fixed the issues with the initial components. If the distributor is good and adjusted properly then fuel is likely the issue.

I changed the battery and it helped some, but now it's saying cylinder 2 misfire and a bad coil. but I put a new coil on when I changed the rotor and cap.

Can you tell me what all the new components you have installed on this truck?

I put new wires, plugs, distributor cap, rotor, coil and ignition control module.

Have you made sure that the distributor does not have any play in it. Loose distributor can give random misfire codes. Other than that it can be the spyder injectors or ECM.
Does it misfire all the time, or it will run good for a while then misfire?

Misfire the whole time it's running.

From everything that you have done so far only item really left is the injectors or a bad ECM.
