EMERGENCY PROBLEM TRANNY OR REAR END OR OTHER????

my truck makes a winding noise through first and second gear and a small point in 3rd gear...only when it runs for a few minutes. I've reved it up and no noise...only when i drive it. i had my transmission flushed new spark plugs and a tune up and it still makes noise. its a 2wd gmc sierra reg cab with 130,000 miles. 5.3 liter v-8 gmc sierra 1500 reg cab

The best way to go after this is to have it up in the air, either on a lift (best) or jackstands, so that you can run it in drive and locate the source. I assume the rear end fluid was checked?
