Front CV axle

I have a 2000 Ford Explorer. Should the passenger CV axle turn freely while in two wheel drive with the front end is jacked up off of the ground?

It should turn pretty freely. If you are feeling some resistance, it probably has something to do with the brakes holding just a little bit. To test, you can use a C-clamp to push the brake caliper piston in just a bit and then put the wheel back on and see if it turns more freely. If so, then that is your issue. If not, then the issue is somewhere else. Probably the hub or axle.
