Do I have to take the tire off to take apart the driveshaft assembly to take the transmission out?

Hi,

Yes, you need to remove the wheel. Then you will see the hub nut. That needs to be removed, along with a few other things, to remove the CV axle.

I attached the directions for removing the axle below. Please review them and let me know if they help or if you have questions.
