Cylinder location

2008 DODGE NITRO

In my service manual it gives a fire in order as 123456, it says 246 left Bank, 135 right bank, i’m confused as to what that means, does it mean 135 on the passenger side or 135 on the driver side when I look online they said on that engine it was turned backwards further confusing me any help you can give would be greatly appreciate it it’s a 2008 dodge nitro 4.0l v6, thanks.

Different engines and makers lay out their engines in different ways. Normally cylinder number one will be the cylinder farthest to the front of the engine.
In the case of your engine the "backwards" would be as compared to other makes. Cylinder one on that engine is the first cylinder on the passenger side. Then behind it are 3 and 5 The driver's side then becomes 2-4-6.
Now if you take that same engine and turn it sideways like in a front wheel drive. The numbers stay the same, so in that case 135 would be the side of the engine facing the firewall and 246 would be the bank facing the radiator.

The image in my service manual is the opposite of the of the image you sent. Does that make a difference?

Same thing, just the one I posted would be if you were standing in front of the car, your image shows the layout from the driver's seat. Note, the arrows showing the direction of the front of the vehicle.

Just don't expect every engine to have the same layout. It can get a bit interesting depending on the make and engine.
