When I start the truck cold it starts up great and can run great as long as I do not turn it off. If I turn it off when it is warm and try starting it again while warm it will not fire on cylinder one. It has new wires, plugs, coils. I have been told vacuum leak, but would a vacuum leak effect all cylinders?

Here is an article that may help you diagnose your misfire:

Performe these tests when the vehicle is warmed up to duplicate the concern. Let us know what the readings are please.
