when getting full service and transmission line replaced, should a mechanic be responsible of refilling the transmission, and how much fluid does transmission hold?

YES they should be responsible! Filling the transmission back up is part of the job. Depends on the transmission, mine holds about 5 qt. You can call your local aout parts store they should be able to tell you
