Removing the charcoal canister would result in emission control fault that would trigger the check engine light. Blocking the pipe from fuel tank would result in the fuel tank not being able to breathe and can result in excessive pressure in fuel tank or vacuum if fuel is being drained too fast. This would result in driveability issues. Why would you want to do that?
The above has nothing to do with the fuel gauge not registering which could be due to wiring fault or a fuel sending unit/meter gauge going bad. You would have to diagnose the cause to determine the rectification method.
