Can you replace a piston in the brake caliper? Its a 4 piston caliper on a 06 tacoma 4WD

You could but usually the cost of all the parts required exceeds the cost of exchanging it for one already rebuilt. You need some special tools also.

What is the difference in a "loaded" or "semi-loaded" caliper? Which do you prefer?

A loaded comes with the pads included and unloaded doesn't. Semi loaded must be some marketing gimmick.
