I am attempting to tighten up the serpentine belt on my 2000 toyota corolla and the reason it was off is because I replaced the alternator. Now if I loosen the bolt for the pulley on the alternator and put the belt on, then tighten it back without messing with the tensioner will that be effective?

no, just release the tension for the belt, install the belt and release the tensioner.
