My 2000 ford Windstar LS has just lost reverse gear, but all other gears are running fine. I there a simple fix for this or something I should try? Or am I in need of a transmission?
My reverse over night slipped but worked and then the next day was gone. There were no warnings. All other gears work fine

This is a Front wheel drive automatic Transmission, does that still the answer for the question? And if so would a Transmission Flush and fill with New filter work?

No, it will not. Beyond a flush. It has an internal failure and will have to be removed, evaluated and most likely re built.
