will a Intake manifold vaccum leak cause a hole to burn in top of piston?

i have a misfiring on #5 cylinder if it is cause by a intake manifold vaccum leak will a hole burn in top of piston on that cylinder if not fix.

If it is bad enough it will cause a lean burn situation that will burn a hole in the top of a piston.
