Hi, I just purchased a R.H exhaust manifold for my 1995 ES300 along with the gasket (3layers welded together). My question is do I use a type of silicone on both sides for the gasket to sit in place and for a proper seal?

No sealers are used on exhaust gaskets. The three layers will compress to fill any voids. A lot of engines don't even use gasket between the manifold and the engine.

I don't recommend using any sealant. If the head and the manifold are true (not warped) the gasket will seal it fine. Just make sure to torque the nuts to 41 ft/lbs.
