Assuming nobody welded your door shut as a prank, the most likely cause is a broken or disconnected linkage to the door release or the release mechanism itself. Is there any resistance in the handles when you try to open the door? If it is loose and doesn't get to a point where it feels like it is "pulling" a little it is not connected. You have to remove the inner door pannel and inspect the linkage to find out. This will also allow you to trip the locking latch mannually to get the door open. Hope this helps.
