I recently had the intake manifold gasket replaced on my two wheel drive vehicle. Since that time it appears to have a transmission slip/clunk when it shifts into overdrive. Is it possible there could be a loose, disconnected or deteriorated vacuum line causing this issue? If so, where is the line located?

The transmission uses no vacuum, but you could still have a vacuum leak. Check hoses etc. Scan for codes auto parts stores does it for free. It may be a coincidental thing that has happened.
