Possible Tune-Up Questions for 2000 Camry

I have a 4-cylinder automatic 2000 Camry that's misfiring in the 3rd and 4th cylinders.

I am not very knowledgeable about cars, but the guys at AutoZone said it probably needs a tune-up.

What all do I need to do and change to fix this, and how would I do it?

Start by changing the spark plugs then go from there.
