the car shakes from the front wheel passenger side when car is picking up speed on road and when you step on the brake to stop.

Sounds like the brake rotor is warped-get it mike out

Is it wise to replace both rotors? And should I have alignment done also?

Good idea on both-I don't think you need realignment if everything is position back correctly.
