Is your firing order correct?

How are you testing for spark?

Have you swapped number six plug wire with another (like number one)?

Pull the cap and see if the contact inside the cap is corroded away.
When was the last time it got a cap/rotor/wires/plugs?

Also, a hairline crack on the distributor cap could cause this. I had one do the same to me. I chased it all around the ignition system, replacing the plug, the wire, cleaning the cap contacts, and so on. Finally I put a new cap on. When I got the old one into the light, I could see the crack in it leading from the number one cylinder (the one that was not firing) towards the center contact. Presto change-o, it purred like a kitten again when I fired it up. So, get a new cap.

Okay, you had two new caps and get spark at all towers except number six?
How and where did you test for the lack of spark? End of the wires? At the plug? With a spark plug or spark tester?
